Effect of Progesterone on Cholecystokinin-induced Amylase Release in Isolated Rat Pancreatic Lobule

Abstract
Since the roles of steroid hormones on pancreatic exocrine secretion are not fully understood, we examined the effect of exogenous progesterone on pancreatic amylase secretion stimulated by cholecystokinin (CCK) using the isolated pancreatic lobules of rats. Pancreatic lobules were incubated with varying concentration of CCK (10 pM~100 nM), and then the mean percentages of total cellular amylase released over a 90-min incubation were subjected to statistical analysis. Estrous stages were determined by vaginal smear. To confirm the roles of exogenous progesterone on the amylase secretion, progesterone (50 mg/kg) dissolved in corn oil were subcutaneously injected to bilateral ovariectomized rats once daily for 2 days. The dose-response curve of CCK in pancreatic lobules prepared from ovariectomized rat was shifted to the right compared to that observed at diestrus, and this response was restored by progesterone treatment. We conclude from the above results that progesterone released from the ovary at diestrus could enhance CCK-induced amylase secretion in the pancreatic lobules of rats.
